About the job Principal Scientist/Associate Director siRNA and ASO Gene Silencing
Summary
We are seeking a highly motivated and skilled scientist to join our team in the development and application of siRNA and ASO technologies for gene silencing. The successful candidate will be responsible for designing, optimizing, and implementing nucleic acid-based strategies to modulate gene expression for therapeutic purposes. This role requires expertise in molecular biology, RNA therapeutics, and gene silencing mechanisms.
Key Responsibilities
Design and Development:
- Design siRNA and ASO molecules targeting specific genes to modulate expression through RNA interference (RNAi) or antisense mechanisms.
- Optimize sequence specificity to minimize off-target effects while maximizing efficacy.
-Mechanism Exploration:
- Investigate mechanisms of action for siRNA.
- Evaluate mRNA degradation pathways, pre-mRNA splicing alterations, and protein translation inhibition.
-Experimental Validation:
- Conduct in vitro and in vivo experiments to validate the efficacy of siRNA/ASO molecules.
- Use molecular techniques such as qPCR, Western blotting, RNA sequencing, and 5 RACE to assess target knockdown efficiency.
- Perform dose-response studies to determine optimal concentrations.
-Therapeutic Applications:
- Collaborate on projects aimed at developing siRNA/ASO-based therapies for diseases such as autoimmune, neuroscience, or metabolic diseases.
- Explore delivery systems (e.g., nanoparticles, liposomes) to enhance cellular uptake of siRNA.
- Investigate chemical modifications of ASOs to improve stability and reduce immunogenicity.
-Data Analysis and Reporting:
- Analyze experimental data to assess on-target and off-target effects.
- Present findings in internal meetings and contribute to patent applications.
-Collaboration:
- Work closely with cross-functional teams including antibody discovery, protein science, biology and pharmacology.
- Stay updated on advancements in RNA therapeutics and contribute to strategic planning.
Qualifications
- Ph.D. degree in Molecular Biology, Biochemistry, Genetics, or a related field with relevant experience.
- Strong understanding of siRNA and ASO mechanisms of actions.
- Hands-on experience with nucleic acid design tools and molecular biology techniques.
- Familiarity with delivery systems for nucleic acids (e.g., lipid nanoparticles).
- Knowledge of therapeutic applications of gene silencing technologies.
Preferred Skills
- Experience with allele-specific RNA silencing strategies.
- Expertise in chemical modifications of oligonucleotides for enhanced stability.
- Knowledge of regulatory considerations for RNA-based therapeutics.
